Abstract:
The present study is interested to demonstratate the renoprotective effect of n-BuOH extract of Centaurea choulettiana Pomel against cisplatin induced nephrotoxicity in mice (Albinos wistar) species. The in vitro study has been condected on the following tests (DPPH °, reducing power, total polyphenols, flavonoids and inhibition of the peroxidation of linoleic acid) had showed that the plant subject of study is endowed with antioxidant properity. On the other hand the plant extract has shown a marked anti-inflammatory effect. Administration of a single dose of cisplatin (8mg/kg bw) intraperitoneally resulted in acute renal deterioration as evidence by the elevation of, creatinine, urea, and renal oxidative stress markers associated with extensive vacuolization of epithelial cell, swelling, desquamation and necrosis. Pretreatment of mice with n-BuOH extract at a dose of (150 mg / kg bw) and vitamin E at a dose of (100 mg / kg bw) for 10 days has dimished the levels of biochemical parameters cited above and MDA, and the restoration of the activity of antioxidant enzymes (CAT, SOD, GPX, and GST) and GSH. Histological protection was clearly observed by reducing the rate of renal MPO. Both biochemical and histopathological results have proved the renoprotective potential of Centaurea choulettiana, which has been able to ameliorate CP- induced and renal dysfunction through its antioxidant power.
