This study aims to provide a scientific basis for an alternative treatment to GFD in celiac disease in adults using the seed of Nigella sativa. Initially we wanted to trace the clinical profile of the disease in eastern Algeria, to achieve this objective a cross-sectional study based on 73 patients was conducted. The second objective was to study the effect of oral administration of Nigella powder on clinical manifestations of the disease by comparing
it with that of gluten-free diet as current treatment of the disease valued in a third part via a study Retrospective range of 35 subjects. The fourth objective of this thesis was to prove the potential of the NS proteolytic protease on gliadin.
The results show that celiac disease in adults in our population is a general disease of young adults, A clean feminine predominance is noted, it is characterized by a variable clinical picture and inconsistent, which may be expressed by a variety of signs intestinal symptoms and extradigestifs mainly represented by chronic diarrhea, abdominal pain, asthenia, weight loss and mucocutaneous pallor.
Oral administration of NS powder at two grams per day over two taken presented a
potential therapeutic effect against the clinical manifestations of celiac disease in adults. The NS anti-fatigue effect in this disease has been proven.
The crude enzyme extract of Nigella sativa seeds presented proteolytic activity of 84.26 UP, with an optimum pH of 1.5 and an optimum temperature of 50 ° C. This protease resisted whatever changes in pH or temperature values. We were able to demonstrate a proteolytic activity of protease Nigella on gliadin. The partially purified Nigella sativa seeds protease presented proteolytic power against gliadin, this was confirmed by analyzing the results of the SDS-PAGE and RP-HPLC. The degradation of gliadin of wheat (Triticum aestivum) was more effective compared to the gliadin of durum wheat (Triticum durum).
It is suggested that Nigella sativa track may represent a therapeutic potential in the
treatment of celiac disease in adults from the point of view of its immunomodulatory effect,anti-asthenic and improving clinical manifestations, of its nutritional value in addition to its protease degrading gliadin.
