dc.description.abstract Gliomas are the largest group of malignant brain tumors in adults. Epidemiological, clinical, translational, and fundamental research have made significant progress in our understanding of these tumors. Nevertheless, the data on the populations of the developing countries remain very scarce, especially because of the rarity of these tumors. The purpose of our study was to develop an epidemiological and genetic profile of patients with glioma in the East-Algerian population. The epidemiological part was based on the records of patients admitted to the radiotherapy-oncology department of the University Hospital Center of Constantine between January 2008 and October 2016, in total 366 records were examined. The genetic part concerned 66/77 patients enrolled at the same hospital between March 2014 and October 2016 as well as 72/82 controls. We analyzed a series of 26/29 polymorphisms identified by GWAS as associated with the risk of gliomas. Our population was young (average age 44.6 years) with a relatively good overall survival (5-year rate of 38.1%). Genetic analysis showed a single polymorphism strongly associated with glioma risk; rs2736100 (5p15.33, TERT). Two other germinal variants gave significant p-values in their heterozygous form; rs6010620 (20q13.33, RTEL1) for association and rs4977756 (9p21.3, CDKN2B-AS1) for protection. The total number of alleles at risk was associated with susceptibility but not with patient survival. Survival was associated with rs3851634 (12q23.33, POLR3B), rs3751667 (16p13.3, LMF1) and rs12076373 (1q44, AKT3). The profile of genetic susceptibility to gliomas in our population may therefore be different from other global populations already described
